In this Journal of Virology® Seminar Series webinar, Alexa Wilson, PhD (Dalhousie University) showcases high‑resolution transmission electron microscopy, confocal microscopy, and spinning disc microscopy to examine how Kaposi’s sarcoma–associated herpesvirus (KSHV) engages the nucleoplasmic reticulum (NR) during lytic replication.
Wilson also presents new advances in live‑cell imaging that enable tracking of individually labeled viral genomes as they enter, move through, and exit the NR—revealing a previously underappreciated pathway for primary envelopment and nuclear egress. Integrated throughout the session, she highlights the dynamic morphology of the NR at ~50,000× magnification, demonstrates cutting‑edge techniques for labeling and tracking herpesvirus capsids, and explains how this updated model of NR‑mediated egress reframes the search for future antiviral strategies targeting early replication steps.
